Purpurfargen følger livet til Celie Johnson og hennes kamp gjennom livet tidlig på 1900-tallet. Filmen begynner da Celie er omlag 17 og føder sitt andre barn, med sin egen far som barnefar. Faren tar barnet fra henne og ber henne om aldri å fortelle noen om det. Tiden går, og den lokale bonden "Mister" kommer for å gifte seg med Celies yngre og penere søster Nettie. Faren nekter å la Nettie gifte seg og gir mannen Celie i stedet. Mister forsømmer og misbruker Celie helt til Nettie vil bo hos dem fordi faren ikke klarer å holde seg unna henne. Da Mister ikke klarer å overtale Nettie, sender han henne avgårde, noe som gjør Celie mer fortvilet enn noensinne. Nettie lover å skrive hvis ikke døden hindrer henne, men Celie får aldri noe brev. Samtidig kommer Misters elskerinne Shug Avery på besøk, blir venn med Celie, og får henne til å forstå at hun er noe mer enn Misters tjener.
